人胰腺癌与端粒酶及其亚单位表达的关系

目的：探讨粒酶活性及其亚单位表达水平与人胰腺癌发生及其临床病理特征的关系。方法：应用聚合酶链反应－银染法（PCR－银染，聚合酶链反应－酶联免疫吸附试验（PCR－ELISA）、逆转录－聚合酶链反应（RT－PCR）及DNA测序等技术，对24例人胰腺癌中端粒酶活性及其亚单位的表达情况进行研究。结果：24例胰腺癌标本中端粒酶活性及hTERTmRNA表达表达阳性率为87．5％、83．3％，而癌旁组织为12．5％，两组差异有显著性（P＜0．05）。hTERTmRNA的表达是人端粒酶活性表达的关键性因素。端粒酶活性的表达水平与肿瘤的分化程度，有无淋巴结的转移及临床分期明显相关。结论：端粒酶参与了胰腺癌的寻生；hTERTmRNA表达水平的上调可在胰腺癌形成过程中起重要作用。端粒酶活性的表达与人胰腺癌的生物学行为及临床病理特征有关，且有助于判断胰腺癌的恶性程度的患者的预后。

Relationship between telomerase and telomerase expression with pancreatic carcinoma

Objective To directly investigate the relationship between the telomerase activity,telomerase subunit expression and pancreatic carcinogenesis,clinical features.Methods Twenty-four cases of pancreatic carcinoma were analyzed for telomerase activity and the expression of subunit by polymerase chain reaction (PCR),PCR-silver staining,PCR-ELISA and DNA sequencing methods.Results Telomerase activity and hTERTmRNA were detected in 24 case.The analysis of pancreatic carcinomarevealed that telomerase activities were positive in 21 of 24 cases (87.5 %),hTERTmRNA in 20 of 24 cases (83.3 %);normal tissues were 12.5 %.There was a significant difference between the two groups (P<0.05).There was obvious correlation between the expression level of telomerase activities and histologic differentiation,metastasis and clinical stage of pancreatic carcinoma.Conclusion The expression of telomerase activity has an relation to carcinogenesis.A strong correlation was found between telomerase activity and hTERTmRNA expression.The up-regulation of hTERTmRNA expression may play a critical role in human carcigenesis.The telomerase activity and subunit level of pancreatic carcinoma were closely correlated with clinical stage of pancreatic carcinoma and may hence be helpful to evaluate the prognosis.
