Abstract:Objective To study the protective mechanism of the ethanolic extract of Alocasia cucullate(EAC)on the spleen of tumor-bearing mice.Methods C57BL/6 mice were subcutaneously inoculated with melanoma B16-F10 cells on the right side of the back,the 6 unmodeled mice were regarded as normal group(normal),and the modeled 30 mice were randomly divided into model group(model),the cyclophosphamide group(CTX)and the low,medium,and high dose ethanolic extract groups of Alocasia cucullate(lEAC,mEAC,hEAC).Six mice of each group were administered continuously for 10 days.At 24 h after the last administration,the mice were killed by cervical dislocation after eyeball blood sampling.The spleen tissue was stripped and weighed to calculate the organ index.The pathological changes of the spleen tissue were observed by hematoxylin-eosin(HE)staining.The spleen of cell apoptosis were detected by TUNEL fluorescence staining.Immunohistochemical detection of B cell lymphoma 2(Bcl-2),B cell lymphoma 2 associated-X protein(Bax),cysteinyl aspartate specific proteinase 3(caspase-3)protein expression levels in spleen tissue,cell counting kit-8(CCK-8)method detect the effect of alcohol extract of taroia acuminata on the growth activity of normal mouse spleen cells.Results The spleen index of model group and hEAC were 5.26±0.94 and 7.78±1.63 respectively,the difference was statistically significant(P<0.01).In immunohistochemistry,the IOD values of Bcl-2 positive cells in spleen tissues of mice in normal group,model group and hEAC group were 8171.34±1210.74,712.79±203.90,7437.72±1342.33;the IOD values of Bax positive cells were 1235.50±302.81,1.14×104±882.91,5241.06±991.12;the IOD values of caspase-3 positive cells were 2197.56±388.14,1.08×104±700.90,3267.04±531.23,the difference between model group and normal group,between model group and hEAC group,the difference were all statistically significant(all P<0.01).Conclusion The EAC can inhibit the abnormal apoptosis of the spleen tissue of tumor-bearing mice in the tumor microenvironment,and has a certain protective effect on the spleen tissue of the tumor-bearing mice.Its mechanism of action is related to the up-regulation of Bcl-2 anti-apoptosis the activity of the protein is related to reducing the activity of Bax pro-apoptotic protein and reducing the expression of caspase-3 protein.
