高压氧对大鼠脑缺血再灌注后碱性成纤维细胞生长因子及其受体表达的影响

摘    要:目的 探讨高压氧对大鼠脑缺血再灌注后神经细胞碱性成纤维细胞生长因子(bFGF)及其受体(FGFR)表达的影响.方法 成年健康雄性Wistar大鼠80只,随机分为模型对照组和高压氧暴露组.2组再各分5组,分别为腩缺血1 h再灌注后1、3、5、7、14 d组,每组8只.应用线栓法制作大鼠左侧大脑中动脉阻塞再灌流模型(MCAO),进行高压氧暴露,免疫组化检测bFGF和FGFR的表达.结果 脑缺血再灌注后1 d,神经细胞即出现bFGF和FGFR的表达,3 d达高峰,后逐渐减弱,至14 d时仍有表达;高压氧暴露组神经细胞bFGF和FGFR表达的变化趋势与对照组相似,但是同一时间点均明显高于对照组.结论 高压氧通过促进bFGF和FGFR表达,可能激活内源性神经保护机制.

Abstract:Objective To study the effects of hyperbaric oxygen(HBO) on the expression of bFGF and FGFR following focal cerebral ischemia reperfusion in rats. Methods The rat middle cerebral artery occlusion reperfusion (MCAO) model was developed by using the intraluminal filament method. Eighty rats were divided into 2 groups: the control group (n=40) and the HBO group (n=40). Then, the 2 groups were further subdivided into 5 groups, i.e. the day 1 reperfusion group, and the day 3 reperfusion group, the day 5 reperfusion group, the day 7 reperfusion group, the day 14 reperfusion group. The animals in these subgroups were all given reperfusion following 1-hour cerebral ischemia. Rats of the HBO group were exposed to hyperbaric oxygen. The expression of bFGF and FGFR were determined by immunohistochemical staining. Results In the control group and the HBO group, the expression of bFGF and FGFR was shown day 1 after ischemia, reached a maximum at day 3, then subsided gradually. In the HBO group, the number of cells was significantly more than those in the control group at the same time point. Conclusions HBO might play an important role in the protection of ischemie neurons by means of over-expression of bFGF and FGFR.
