| 摘 要： | 采集6月龄河南良杂猪的脾脏,分离淋巴细胞后直接提取总RNA,进行反转录-聚合酶链反应(RT-PCR)扩增,扩增产物进行T-A克隆、测序,获得了河南良杂猪IL-18基因成熟蛋白序列.序列测定表明,pIL-18成熟蛋白基因核苷酸长度为474 bp,编码157个氨基酸.将该基因片段克隆到大肠杆菌表达载体pQE30,构建重组质粒pQE-IL18,所获重组质粒经过酶切、测序鉴定,证实含有目的片段,且连接、构建正确.

| Abstract: | Porcine mature protein IL-18 gene was amplified by RT-PCR from porcine splenocyte total RNA extracts and then was cloned and sequenced.The result suggested that the sequence of porcine pIL-18 gene consisted of 474 bp,which encodes 157 amino acids.Then,mature protein gene was subcloned into the prokaryotic expressing plasmid vector pQE30 to construct and recombine the pQE-IL18 plasmid.The obtained recombinant plasmid was proved to contain the desired fragments,and be joined and constructed correctly by digestion and sequence identification. |
