灯盏乙素抑制LPS诱导BV2细胞株炎症细胞因子表达的研究

摘    要:目的研究灯盏乙素对脂多糖(lipopolysaccharide,LPS)诱导BV2细胞株炎症细胞因子表达的影响,探讨灯盏乙素抗炎作用机制。方法通过荧光素酶报告基因检测核转录因子NF-κB活性,并利用LPS刺激使BV2细胞和通过RT-PCR和蛋白免疫法检测TNF-α和IL-1β的mRNA及蛋白含量,研究灯盏乙素的抗炎作用。实验分为正常组、LPS刺激(1μg/ml)的模型组、灯盏乙素组(0.1、1、10和100μg/ml)。结果灯盏乙素预处理(1~100μg/ml)可抑制LPS引起pNF-κB 293的核转录因子NF-κB激活(P<0.05或P<0.01);灯盏乙素预处理(1~100μg/ml)能够抑制LPS引起BV2的TNF-α和IL-1βmRNA和蛋白含量升高(P<0.01)。结论灯盏乙素具有抗炎作用,其抗炎作用与抑制核转录因子NF-κB、抑制炎症细胞因子表达有关。
