PROTEOME ANALYSIS IN BILIARY DIFFERENTIATION OF WB-F344 CELLS BY ITRAQ TECHNOLOGY

Abstract:Objective To screen for the differentially expressed proteins during WB-F344 cells biliary differentiation by iTRAQ technique.Methods The WB-F344 cells were cultured and treated in sodium butyrate.The total protein at 0,2,4,6d during the differentiation was extracted and analyzed by iTRAQ technology coupled with mass spectrometric analysis.Results A total of 67 significant protein spots were found by mass spectrometric analysis,among which,33 spots were up-regulated and 34 spots were down-regulated in WB-F344 cell differentiation.Conclusion This study indicated iTRAQ is a useful technologic method in proteomic study of cell differentiation and the results provide the experimental foundation for elucidating molecular mechanism of WB-F344 cell differentiation.
