氯化镧对仔鼠海马BDNF表达的影响

摘    要:为了探讨氯化镧对仔鼠海马组织BDNF表达的影响。将健康成年雌性Wistar大鼠32只,随机分为对照组(饮用蒸馏水);低剂量染镧组(饮用含0.25%LaCl3的蒸馏水溶液)、中剂量染镧组(饮用含0.5%LaCl3的蒸馏水溶液)和高剂量染镧组(饮用含1.0%LaCl3的蒸馏水溶液),每组8只动物。然后将雌雄大鼠按1∶1同笼进行交配,发现阴栓或阴道分泌物镜检有精子者记为妊娠第0 d,孕鼠单笼饲养并自由摄水。子代大鼠在断乳前经由母体血循环和吸吮母乳染镧,断乳后则通过自行饮水摄入镧,至断乳后1个月结束。染毒结束后取海马组织,利用实时定量PCR的方法检测仔鼠海马组织内BDNF mRNA表达,利用western blot的方法测量仔鼠海马组织内BDNF蛋白的表达。研究发现高剂量组仔鼠海马BDNF mRNA表达显著低于中剂量、低剂量染镧组及对照组(P〈0.05),中剂量组仔鼠海马BDNF mRNA表达显著低于低剂量染镧组及对照组(P〈0.05),低剂量染镧组仔鼠海马BDNF mRNA表达显著低于对照组(P〈0.05);对照组仔鼠BDNF蛋白表达的灰度值为(0.81±0.09)、低剂量染镧组BDNF蛋白表达的灰度值为(0.68±0.07)、中剂量染镧组BDNF蛋白表达的灰度值为(0.54±0.09)、高剂量染镧组BDNF蛋白表达的灰度值为(0.38±0.05),仔鼠海马BDNF蛋白表达与染镧剂量负相关(P〈0.05)。结果表明亚慢性氯化镧暴露大鼠海马组织中BDNF表达降低,可能是氯化镧对神经系统毒性损伤的机制之一。

Abstract:To investigate the effects of lanthanum chloride(LaCl3) on BDNF expression in hippocampus tissues of offspring rats.Healthy adult female Wistar rats were randomly divided into four groups,including control group(distilled water),low-dose group(drinking water containing 0.25% LaCl3 solution in distilled water),middle-dose group(drinking water containing 0.5% LaCl3 solution in distilled water) and high-dose group(drinking water containing 1.0% LaCl3 solution in distilled water),n = 8.Occurrence of copulatory plug was used to indicate successful mating and day 0 of pregnancy,pregnant rats were single caged and drank water freely.The pups were exposed to LaCl3 by parental lactation for postnatal 3 weeks and then continue orally administrated with 0%,0.25%,0.50%,and 1.0% LaCl3in distilled drinking water for 1 month,respectively.Hippocampal tissues were taken after the end of exposure,and the levels of brain derived neurotrophic factor(BDNF) in the hippocampi of pups were determined by real-time quantitative PCR and western blot.The results from this study found that BDNFmRNA expression in the hippocampus of high-dose group was significantly lower than in the middle-dose group,low-dose group and the control group(P 0.05),BDNFmRNA expression in the hippocampus of middle-dose group was significantly lower than in the low-dose group and control group(P 0.05),BDNFmRNA expression in the hippocampus of low-dose lanthanum was significantly lower than in the control group(P 0.05); The IDV of BDNF protein expression in control group was 0.81 ± 0.09,the IDV of BDNF protein expression in low-dose group was 0.68 ± 0.07,the IDV of BDNF protein expression in middle-dose group was0.54 ± 0.09 and the IDV of BDNF protein expression in high-dose group was 0.38 ± 0.05,BDNF protein expressions in hippocampi were negatively correlated with the dose of lanthanum(P 0.05).It suggested that chronic exposure to LaCl3 reduced the expressions of BDNF,and which may be one of toxic injury mechanisms in the nervous system of LaCl3.
