人骨肉瘤细胞凋亡及p53、p16及bcl-2的表达研究

目的　研究人骨肉瘤细胞凋亡及p5 3、p16、bcl- 2的表达情况 ,并初步探讨其临床及生物学意义。方法　采用原位DNA末端标记法及免疫组化方法检测了 39例骨肉瘤细胞凋亡及p5 3、p16、bcl- 2的表达情况 ,并统计其阳性反应率。结果　本组病例中细胞凋亡阳性率 6 1 5 % ,p5 3呈阳性反应 2 9例 ,阳性率 74 3% ,p16呈阳性反应 2 7例 ,阳性率 6 9 2 % ,bcl- 2呈阳性反应 7例 ,阳性率 19%。结论　骨肉瘤组织中有p5 3、p16、bcl- 2基因的高表达 ,与肿瘤的预后相关。在骨肉瘤中细胞凋亡是一种自然现象 ,其发生情况各不相同 ,研究初步提示了其与预后相关 ,但应与p5 3、p16、bcl- 2蛋白的表达相结合而综合判断。

Apoptosis and expression of p53,p16 and bcl-2 in osteosarcoma

Objective TO determine the expression of apoptosis,p53,p16 and bcl-2 in ostessarcoma and to evaluate preliminary it's significance in clinic and biology.Methods A series of 39 osteosarcoma was analyzed for the expression of apoptosis by 3-end-labeling method of DNA in tissue section,and the expression of p53,p16 and bcl-2 by immunohistochemical technique.The indexes of apoptosis,p53,p16 and bcl-2 were respectively the percentages of apoptosis cells or p53,p16 and bcl-2 positive cells.Result Apoptosis occurred in 61 5% of osteosarcoma,the positive rate of p53 is 74 3%(29/39),p16 is 69 2%(27/39),and bcl-2 is 19%(7/39).Conclusion There were p53,p16 and bcl-2 high expression in osteosarcoma,and the high expression associated with prognosis.The apoptosis was a spontaneous phenomenon in osteosarcoma,and the expression was different from each other.The apoptosis associated with prognosis,and it is had better to judge with the expression of p53,p16 and bcl-2.
