海参肠道β-1,3-葡聚糖酶的提取条件及其酶学性质

摘    要:对海参肠道β-1,3-葡聚糖酶的提取条件及酶学性质进行了研究.结果表明,最佳提取条件为:pH6.0的磷酸盐浸提液0.05 mol/L,NaCl浓度为0.05 mol/L,缓冲液体积与海参肠质量之比为4∶1,硫酸铵饱和度为80%;该酶最适反应条件为:温度40℃,pH6.0;该酶在10~40℃,pH5.0~6.5酶活力稳定.Mn2+对酶具有一定的激活作用,Cu2+、Ca2+、Mg2+、Fe2+、Zn2+、Ba2+、K+、Ag+对酶存在不同程度的抑制,其中Cu2+对酶的抑制作用最强.

Abstract:Extraction and properties of β-1,3-glucanase from the gut of sea cucumber were investigated in this paper.The optimum extraction condition was obtained as follows: phosphate buffer(pH 6.0,0.05 mol/L,0.05 mol/L NaCl): gut=4∶1,saturation of(NH4)2SO4 was 80%.The optimum temperature and pH of the enzyme were 40 ℃ and pH 6.0.The crude β-1,3-glucanase was stable at pH 5.0-6.5.The enzyme was activated by 0.05 mol/L Mn2+ and inhibited by Cu2+,Ca2+,Mn2+,Mg2+,Fe2+,Zn2+,Ba2+,K+ and Ag+,in which Cu2+ was strongest inhibitor.
