摘    要:运用紫外、微波复合诱变技术,对高效石油降解菌K05—2—11进行改良,根据致死率曲线,选择紫外诱变时间为100s、微波诱变时间为455。经过连续4次紫外诱变,得到紫外诱变改良菌UV2;将其作为微波诱变的出发菌株,经2次微波诱变,最终得到遗传稳定的高效耐高温石油降解菌UM2—6。比较K05—2—11和UM2—6的温度耐受性发现,UM2—6的耐高温性能良好,在55℃的高温下仍具有27.42%的石油降解率,其最高石油降解率出现在45℃,达到48.72%,较K05—2-11提高20.81%,改良效果明显。

Abstract:.UV-Microwave mutagenesis technique was used to improve the efficient petroleum degradation strain KO5-2-11. Based on the lethal rate curves, the mutagenesis time of UV and microwave were determined as 100 s and 45 s, respectively. After the accumulation of UV mutagenesis for four times, the improved strain UV2 was achieved,which could be used as the original strain for microwave mutagenesis. After twice micro- wave mutagenesis,an efficient petroleum degradation strain UM2-6 with high genetic stability and thermosta- bility was finally obtained. The thermostablity comparison between KO5-2-11 and UM2-6 showed that under the high temperature of 55 ℃, UM2-6 still had a petroleum degradation rate up to 27.42 % with high heat tolerance,and the maximum petroleum degradation rate was 48.72% at 45 ℃ ,20.81% higher than that of KOS-2- 11,showing obvious effect of improvement.
