缺氧对ECV-304细胞CASK表达及细胞内分布的影响

摘    要:目的 了解缺氧后ECV-304细胞钙/钙调蛋白依赖性丝氨酸蛋白激酶(calcium/calmodulin-dependent serine protein kinase, CASK)蛋白表达及细胞内分布的变化.方法 将培养的ECV-304细胞置于缺氧(5%O2) 条件培养1、3、6、12 h,用Western blot方法检测CASK表达的变化,用免疫组化技术观察CASK在细胞内分布情况.结果 ECV-304细胞的CASK表达随缺氧时间延长而逐渐增高,3 h开始增加,6 h达高峰,12 h下降但仍高于正常.免疫组织化学染色的结果显示,随着缺氧时间的延长,细胞染色加深、并出现CASK向核膜聚集的现象.结论 缺氧可以影响CASK的蛋白表达和细胞内分布,提示CASK可能是一个新的值得关注的缺氧相关蛋白.

Abstract:Objective To explore the changes of CASK(calcium/calmodulin-dependent serine protein kinase) expression and cellular distribution in ECV-304 cells under hypoxia.Methods ECV-304 cells were exposed to 5%O2 for 1,3,6,12 h.The CASK protein level was detected by Western blot and the cellular distribution was observed by using immunohistochemical staining.Results After hypoxia exposure,the expression of CASK protein was increased in a time-dependent manner.The elevation began at 3 h after hypoxia,peaked at 6 h,then decreased but still above normal level at 12 h.The cellular distribution of CASK also changed,with a tendency of translocation from cytoplasm to nuclear membrane.Conclusion Hypoxia may upregulate CASK expression in ECV-304 cells and could also alter its cellular distribution.
