青白通痹方药半仿生提取工艺条件优选研究

摘    要:目的:优选青白通痹方药半仿生提取的工艺条件。方法:在饮片规格、煎煮温度、用水量、滤过、浓缩等基本提取条件相同的情况下,对第一煎水pH(A)、第二煎水pH(B)、第三煎水pH(C)和煎煮时间h(D)4个主要因素各3个水平进行考察。采用L9(3^4)正交表,综合评判指标选取青藤碱、芍药苷、甘草次酸、分子量≤1000Da提取物、总生物碱和干浸膏,优选青白通痹方药半仿生提取的工艺条件。结果:3次提取用水的pH值依次为2.0、7.0、9.0;3煎总计时间为5.0h。结论:考虑到大生产的实际情况,确定青白通痹方药的半仿生提取的工艺条件为3次提取用水的pH值依次为2.0、7.0、9.0;提取时间依次为2.0h、1.50h、1.50h。

Optimization of semi-bionic extraction technical conditions for Qingbai Tongbi decoction

Abstract:Objective:To optimize the technical conditions of semi-bionic extraction(SBE) for Qingbai Tongbi decoction.Methods:3 levels of 4 main factors of the first decoction pH (A),the second decoction pH (B),the third decoction pH (C) and decoction time h (D) were examined under the same conditions as the basic extraction conditions such as the size of the pieces,the boiling temperature,the water consumption,the filtration,and the concentration.Using L9 (3^4) Orthogonal design and taking sinomenine,paeoniflorin,Glycyrrhetinic acid,≤1000Da extraction,total alkaloid and dry extracts as comprehensive evaluation indexes,the result was comprehensive evaluated to optimize SBE technical conditions for Qingbai Tongbi decoction.Result:The pH values of 3 times of extraction was 2.0、7.0、9.0 respectively,and the total extracting time was 5.0h.Conclusion:Combinating manufacturing fact,the pH values of thrice extractions is 2.0、7.0、9.0 respectively,and the extracting time is 2.0 h、1.50 h、1.50 h in turn.
