大剂量甲基强的松龙联合MK-801治疗大鼠急性脊髓损伤的实验研究

目的观察大剂量甲基强的松龙（MP）联合MK-801（地卓西平马来酸盐）对大鼠急性脊髓损伤的治疗效果。方法采用Allen的重物坠落法建立大鼠急性脊髓损伤模型，将72只致伤大鼠一次性随机分为4组，每组18只大鼠（其中每组又包括24h、72h、15d三个亚组，每个亚组6只大鼠）。对照组伤后0．5h腹腔注射5mL生理盐水；MP组伤后0．5h腹腔注射甲基强的松龙30mg／kg体重；MK-801组伤后0．5h腹腔注射2．5％MK-801（地卓西平马来酸盐）5mg／kg体重；联合组伤后0．5h腹腔注射甲基强的松龙和MK-801（剂量、浓度同前）。分别在伤后24h、72h、15d处死动物，观察受损部位脊髓的病理形态和细胞凋亡，其中15d组分别于第1、3、5、7、10、15天做Gales评分和斜板试验。结果甲基强的松龙、MK-801及联合组治疗大鼠急性脊髓损伤，在Gales评分、斜板试验及病理形态上、细胞凋亡检测与对照组相比都有很大提高，但三个治疗组在上述方面却处于同一水平。结论甲基强的松龙、MK-801及两药联合对大鼠急性脊髓损伤的治疗作用在目前的实验条件下无明显差异。

Combined Treatment of Acute Spinal Cord Injury with Methyl Prednisolone and MK-801:An Experimental Study

Objective To observe the therapeutic efficacy of methyl prednisolone combined with MK-801（Dizocilpine Maleate） in rats with acute spinal cord injury. Methods Allen, sWD spinal cord injury rat model was constructed. Seventy-two rats were divided into 4 groups randomly. ： Eighteen rats per group（quorum per group contain Twenty-four hours group.Seventy- two hours group.Fifteen Days group three subgroup,per subgroup six rats）. Control group：Peritoneal injection 5mL physiological saline 0.5 hour after injuried;MP group：Peritoneal injection methylprednisolone 30mg/kg body weight 0.5 hour after injuried; MK-801 group：Peritoneal injection 2.5% MK-801 （Dizocilpine Maleate）5mg/kg body weight 0.5 hour after injuried;combine group：Peritoneal injection methylprednisolone and MK-801 0.5 hour after injuried （dosage.density ditto）. The rats were terminated at the 24 th hour.the 72 th hour and on the 15 th day after trauma, The pathomorphology and apoptosis of the injured site of the spinal cord were observated,cujus 15 days group dis- cem at 1th、3th.5th.7th.10th.15th day commit motor function score and inclined plane test. Results The groups treated with methylprednisolone.MK-801 and a combination of both after acute spinal card injury were compared with control group, significantly enhanced ethological score.inclined plane test and pathomorphology.apoptosis detection . however,there was no obvious differences among the three treated groups. Conclusion The administration of methylprednisolone or MK-801 alone or in combination has not different efficacy obviously existing experiment condition for the treatment of acute spinal cord injury.
