摘    要:目的:采用HPLC法测定武当地区不同品种茶叶及其他地区茶叶的咖啡因和表没食子儿茶素没食子酸酯的含量。方法:以Luna-C18柱(250 mm×4.6 mm,5μm)为色谱柱,流动相为乙腈-0.2%磷酸水溶液,梯度洗脱,流速为0.75 ml·min^-1,检测波长为272 nm,柱温为25℃。结果:咖啡因和表没食子儿茶素没食子酸酯的线性范围分别为10.21~102.10μg·ml^-1(r=0.9994)、16.21~162.10μg·ml-1(r=0.9991),平均回收率分别为98.67%(RSD=1.86%,n=6),98.19%(RSD=2.28%,n=6)。结论:该方法用于武当地区及其他地区茶叶的咖啡因及表没食子儿茶素没食子酸酯含量的快速测定,具有简便、稳定性好、准确度高等特点,可以作为茶叶中咖啡因及表没食子儿茶素没食子酸酯含量的测定。

Abstract:Objective:To determine the contents of caffeine and epigallocatechingallate in different varieties of tea in Wudang and other regions by HPLC.Methods:The chromatographic column was a Luna-C18 column(250 mm×4.6 mm,5μm).The mobile phase was acetonitrile-0.2%phosphoric acid aqueous solution with gradient elution at the flow rate of 0.75 ml·min^-1.The detection wavelength was 272 nm.The column temperature was 25℃.Results:The linear range of caffeine and epigallocatechingallate was 10.21-102.10μg·ml-1(r=0.9994)and 16.21-162.10μg·ml^-1(r=0.9991),respectively.The average recovery was 98.67%(RSD=1.86%,n=6)and 98.19%(RSD=2.28%,n=6),respectively.Conclusion:The method is simple,stable and accurate,which can be used for the determination of caffeine and epigallocatechingallate in tea from Wudang and other regions.
