高脂血症引起小鼠内脏的基因特异性变化

摘    要:通过给小鼠喂食高脂饲料建立小鼠高脂血症模型,并测定内脏中一系列相关基因的表达,为解明高脂血症形成的分子机理提供一定的理论基础.11只6周龄雄性ICR小鼠,随机分成两组:一组为基础饲料组(BD),另一组为高脂饲料组(HFD).实验结果表明:对比BD,HFD血清Tc,LDL-C,VLDL和HDL-C水平有显著上升(p<0.01),而Tg水平明显下降(p<0.05).在肝脏中,Sod基因和Gpx1基因表达显著下降,而ID1和LDL-r表达显著上升(p<0.05).心脏中,HFD的LPL基因表达明显上升(p<0.05).结果表明,上述基因表达水平的变化可能与高脂血症在生理方面带来的不良作用有关.

Abstract:The aim is to establish a mice hyperlipidemia model that induced by high-fat diet and investigate the expression levels of genes that might be involved in important physiological aspects related to the bad influence of the hyperlipidemia process.Eleven four-week-old ICR male mice consumed a basic diet(BD) for five days before they were randomized into two groups.One group was fed BD as a control group(BD) and the other was fed a fat-supplied BD as a high-fat diet group(HFD).On day 34,blood and tissues were collected.Combined with the analysis of total cholesterol(Tc),triglycerides(Tg),HDL-C,LDL-C and VLDL levels in serum by standard methods,the expression of gene Sod1,Sod2,Gpx1,Gpx2,LDL receptor(LDL-r),HMG-CoA reductase(HMG-red),cholesterol 7-alpha-hydroxylase(CYP7A1),inhibitor of DNA-binding-1(Id1),Id3,FAT/CD36 and LPL in different tissues,such as liver and heart,were measured by real-time quantitative RT-PCR.Compared with BD,HFD showed a significantly increase in Tc,LDL-C,VLDL and HDL-C levels(p<0.01) and a significantly decrease in Tg level(p<0.05) in the serum.The expression of gene Sod and Gpx1 in liver was significantly reduced,while the level of Id1 and LDL-r mRNA was greatly increased(p<0.05).Moreover,HFD showed a significantly increase in LPL expression in heart(p<0.05).The present study indicates that the change in the above gene expression may account for the hyperlipidemia-brought bad consequence in physiological aspects.
