摘    要:目的:分析尼美舒利(Nim)对5-氟尿嘧啶(5-Fu)在大鼠体内药代动力学参数的影响.方法:用反相高效液相色谱法精密检测血浆中5-Fu的浓度,研究单用5-Fu及联用Nim后的药代动力学变化.结果:联用Nim后,5-Fu的药时曲线下面积、最大血药浓度增大,清除率下降、达峰时间延迟、消除相半衰期增加.结论:联用Nim后可使5-Fu进入血循环总药量增加,在体内吸收减慢、消除延缓,作用时间延长.为两药联用后人体药代动力学研究提供参考.

Abstract:Objective: To study the effect of nimesulide (Nim) on the pharmacokinetics of 5-fluorouracil (5-Fu) in rats. Methods:The concentration of 5-Fu in the plasma of rats was detected with reverse phase high performance liquid chromatography (RP-HPLC). Results:After administration of Nim, the area under the curve (AUC), maximum plasma concentration (Cmax), half life of elimination, and total amount of 5-Fu in blood circulation increased, and the clearance rate (Clr) and peak time (Tpeak) decreased. Conclusion:Compared with administration of 5-Fu alone, administration of 5-Fu combined with Nim could increase the total amount of 5-Fu in blood circulation, slow down the absorption of 5-Fu, and prolong the action time of 5-Fu. This study provides a good reference for combined administration of 5-Fu and Nim in the pharmacokinetic study.
