摘    要:目的 建立测定四乙酰葛根素的含量及有关物质的超高效液相色谱法.方法 采用Agilent XDB-C18色谱柱(4.6 mm×250 mm,5μm),流动相:乙腈-水(45∶55),流速:1.2 mL·min-1,检测波长:250 nm,柱温:30℃.结果 四乙酰葛根素的浓度在0.020 76~0.186 8mg·mL-1与峰面积线性关系良好(A=1.287×104 C-24.55,r=0.999 9,n=7),检测限为6.65 ng ·mL-1(S/N=3);平均加样回收率为99.9% (RSD=1.4%,n=9);有关物质含量为1.02%~1.28%.结论 该方法简便、准确,可用于四乙酰葛根素的含量及有关物质的测定.

Abstract:Objective To establish a UPLC method to determine the content and related substances of tetra-acetylated puerarin.Methods Agilent XDB-C18 chromatographic column(4.6 mm×250 mm,5 μm)was used and the mobile phase consisted of acetonitrile-water(45∶55)with a flow rate of 1.2 mL·min-1.The UV detection wavelength was 250 nm.The column temperature was 30 ℃.Results The calibration curve was linear over 0.020 76-0.186 8 mg·mL-1(A=1.287×104C-24.55,r=0.999 9,n=7).The detection limit was 6.65 ng·mL-1(S/N=3).The average recovery was 99.9%(RSD=1.4%,n=9).The content of related substances of tetra-acetylated puerarin was over 1.02%-1.28%.Conclusion The method is simple,accurate,and applicable for the quality control of tetra-acetylated puerarin.
