摘    要:目的优选白屈菜中总生物碱的最佳提取工艺。方法采用酸性染料比色法以总生物碱得率为评价指标,首先采用Plackett-Burman实验设计筛选出影响白屈菜总生物碱提取工艺的3个主要因素为乙醇体积分数、料液比、提取次数,在此基础之上,再通过Box-Behnken响应面分析法优化得到影响白屈菜中总生物碱的最佳提取工艺条件。结果最佳提取工艺为:乙醇体积分数为85%,提取时间60 min,提取温度70℃,粉碎程度为40目,料液比为1∶15,提取次数为3次。在此试验条件下,白屈菜中总生物碱得率可达1.92%,与理论预测值1.95%基本一致。结论本试验优选得到的提取工艺合理可行,所得总生物碱得率较高。

Abstract:Objective To optimize the best technique for extracting the total alkaloids from Baiqucai (Herba Chelidonii,Greater Celandine).Methods The yield of the total alkaloids was taken as review index by using acid dye colorimetry,and Plackett-Burman design (PBD) test was applied at first for screening 3 main factors influencing the technique of extracting the total alkaloids from Baiqucai and they were ethanol volume fraction,solid-liquid ratio and extracting times.Then the best technologic conditions for extracting the total alkaloids from Baiqucai were determined by applying response surface analysis (RSA) of Box-Behnken design (BBD) test.Results The best extracting technique was as follows:ethanol volume fraction was 85%,extracting duration was 60 min,extracting temperature was 70℃,degree of grinding was 40 meshes,solid-liquid ratio was 1:15,and extracting times were 3 times.In these conditions,the yield of the total alkaloids from Baiqucai was up to 1.92%,which was basically the same as theoretical prediction (1.95%).Conclusion The extracting technique optimized in this test is reasonable and feasible,and can get higher content of the total alkaloids.
