| 摘 要： | 目的研究氰戊菊酯对C6胶质细胞增殖的影响。方法以3.16、10、31.6和100 mg/L的氰戊菊酯对C6胶质细胞进行染毒,通过四噻氮唑盐(MTT)比色法观察其在不同时间点(24、48、72 h)对细胞增殖的影响;利用B rdU掺入法反映其对细胞DNA合成的影响;使用流式细胞术检测其对细胞周期的作用。结果氰戊菊酯对C6胶质细胞增殖有明显抑制作用,且呈现剂量、时间依赖性;可抑制C6胶质细胞DNA合成;可阻滞C6胶质细胞从G0/G1期进入S期。结论氰戊菊酯能够抑制C6胶质细胞增殖,其抑制作用可能是使细胞周期阻滞于G0/G1期,减少细胞DNA合成。

| Abstract: | Objective To investigate the effect of fenvalerate on cell proliferation of C6 glioma cell. Methods Dose-and time-dependent inhibitory effects of fenvalerate on C6 cell proliferation were deceted using MTT assay. DNA syn-thesis was determined by BrdU incorporation followed by immunocytochemistry staining. Cell cycle stage was deter-mined by FASC. Results Fenvalerate inhibited the proliferation of C6 glioma cell in a dose-and time-dependent manner, and inhibited the DNA synthesis. The cytometry confirmed that fenvalerate could arrest the C6 glioma cell in G0/G1 stage. Conclusion Fenvalerate can inhibit C6 cell proliferation by blocking the cells enter into S stage. |
