小鼠心脏冠状动脉毛细血管内皮细胞的培养对研究其心功能的意义

摘    要:目的:培养小鼠心脏冠状动脉毛细血管内皮细胞,探讨其对研究心功能具有的意义。方法:采用Jian-MeiLi的方法,从ICR小鼠心室肌获得组织,用胶原酶、胰酶和DNase进行消化,通过离心、种植和培养及经VIII因子、CD34相关抗体进行免疫组化鉴定。结果:培养的细胞呈单层贴壁生长,细胞初为圆形,伸展后为多角形和菱形,五六天呈不规则“铺路石样”征象,CD34相关抗体免疫组化鉴定为阳性。结论:体外培养小鼠心脏冠状动脉毛细血管内皮细胞的方法简单、易行,对其心功能的研究具有重要意义。

Abstract:AIM:To culture microvascular endothelial cells from mouse heart,and to assess its significance in studying heart function. METHODS:Mouse heart coronary microvascular endothelial cell was developed and generated according to Jian Mei Li et al.Microvascular endothelial cells were isolated from devitalized mouse ventricular tissue after sequence digestion with collagenase,trypsin and Dnase.Coronary microvascular endothelial cells were separated from cardiac myocytes and other cells by differential centrifugation,planting and culture.The microvascular endothelial cells obtained from above procedures were judged by VIII factor and CD34 related antibody using immunohistochemisty evaluation. RESULTS:At 5,6 d, the cultured cells displayed monolayer anchoring growth and looked like irregular "cobblestone"symbol(round extension to polygon and rhombus in the initial period).The CD34 related antibody showed positive by immunohistochemisty evaluation.
