滇南小耳猪与云南野猪SLA-DQA基因的多态性比较

摘    要:目的 分析滇南小耳猪与云南野猪(S.s.silvianus) SLA-DQA基因不同酶切位点的基因型多态性以及等位基因的多态性,检验这些酶切位点上的基因频率是否达到Hardy-Weinberg平衡.方法 采用EcoRⅠ和AluⅠ两种限制性内切酶对滇南小耳猪和云南野猪SLA-DQA第1和第2内含子部分序列以及完整的外显子2进行PCR-RFLP分析.结果 在EcoRⅠ与AluⅠ酶切位点上均得到3种基因型,在这两个酶切位点上版纳系有9种组合基因型,明光系有8种组合基因型,而野猪仅发现4种组合基因型.经x2适合性检验表明:滇南小耳猪SLA-DQA外显子2在EcoRⅠ与Alu Ⅰ酶切位点上均未达到Hardy-Weinberg平衡状态(P<0.05),但明光系和云南野猪在两酶切位点均达到了平衡状态(P>0.05).结论 滇南小耳猪SLA-DQA在EcoRⅠ与AluⅠ酶切位点均表现为中度多态,云南野猪在EcoRⅠ为中度多态,在Alu Ⅰ酶切位点则为低度多态;滇南小耳猪的HardyWeinberg不平衡主要来自版纳系小耳猪.

Abstract:Objective Analyzed genotype and allelic polymorphism of SLA-DQA gene in Diannan small-ear pig and Yunnan wild boar (YWB, S. s. silvianus) to detect whether the genotype frequency of restriction enzyme sites was fit with Hardy-Weinberg equilibrium. Methods PCR-RFLP was used to analyze the partial sequences of intron l and intron 2 and the complete sequence of extron 2 of SLA-DQA in Diannan small-ear pig(DN) and Yunnan wild boar. Result Both EcoR I and Alu I digested sites had three kinds of genotypes, There were nine composite genotypes in Banna pig(BN) and eight in Mingguang pig(MG), but only four composite genotypes in Yunnan wild boar. The ~2 test value of genotypes at both EcoR I and Alu I sites in Diannan small-ear pig wasn't fit with Hardy-Weinberg equilibrium (P〈0.05), versus in Mingguang pig and Yunnan wild boar was fit with Hardy-Weinberg equilibrium, respectively (P〉0.05). Conclusion Genetic polymorphism of SLA-DQA at both EcoRand Alu I sites in Diannan small-ear pig is moderate, at EcoR I site in Yunnan wild boar is moderate versus at Alu I site in Yunnan wild boar is low; the disequilibrium of Hardy-Weinberg in Diannan small-ear pig is majorly resulted from Banna pig.
