沙漠枯草芽孢杆菌 PJS 产生的环脂肽类抗生素研究

摘    要:目的 分离鉴定枯草芽孢杆菌PJS发酵液中的环脂肽类抗生素PJS-0。方法 采用溶剂萃取、薄层色谱(TLC)、高效液相色谱法(HPLC)等分离方法 对菌株PJS发酵产物的活性组分PJS-0进行分离;并通过紫外(UV)、质谱(MS)和核磁共振波谱(NMR)联合分析对其结构进行鉴定。结果 从塔克拉玛干沙漠来源植物内生枯草芽孢杆菌PJS的发酵液中分离得到一个环脂肽类抗生素:PJS-0,经结构鉴定为Mojavensin A。结论 本文首次从陆生芽孢杆菌中分离得到Mojavensin A,并首次用ESI^+-MS/MS对其氨基酸序列进行鉴定。

Abstract:Objective To isolate and identify compound PJS-0 from the fermentation broth produced by Bacillus subtilis strain PJS. Methods Compound PJS-0 was isolated and purified by using solvent extraction, TLC and HPLC. UV, MS and NMR were analyzed to elucidate the structure of PJS-0. Results Compound PJS-0 was obtained from the fermentation broth produced by Bacillus subtilis strain PJS isolated from Taklamakan Desert, and structurally identified as Mojavensin A, acyclic lipopeptide group antibiotic. Conclusion Mojavensin A is discovered from the fermentation broth of terrestrial-derived Bacillus subtilis strain PJS for the first time and amino acid sequence of Mojavensin A is also analyzed by the tandem mass spectrometry method (ESI^+-MS/MS) for the first time.
