| 摘 要： | 针对虎杖乙醚组分复杂体系的分离,借助于CSASS软件,选用相同的起始体积分数〔φ(甲醇)=10%〕、不同的强溶剂浓度梯度、最终达到相同体积分数〔φ(甲醇)=100%〕的5次简单的线性梯度,快速获取了虎杖乙醚组分中44个峰的保留值方程。同时利用色谱峰自动曲线拟合方法获取了各色谱峰的峰形参数,用分离峰的峰形参数σ、W1/2与tR*线性回归获取了峰形规律,并且用于对谱图的仿真。在此基础上,应用移动重叠分辨分离图和谱图仿真技术,发展了计算机辅助的虎杖乙醚组分的高效液相色谱全局优化方法。在优化条件下,虎杖乙醚组分的HPLC分析可在60 min内完成,当中的常量成分都能够得到较好的分离度。

| Abstract: | For the separation of complex system of ether fraction of P.cuspidatum,CSASS software was used.The LC retention value equation of ether fraction of P.cuspidatum was obtained correctly and quickly by using five linear gradients,in which CB was started with 10% and ended with 100% in different time intervals.Based on the technology of automatic curve fitted,the peak shape parameters and peak shape law in ether fraction of P.cuspidatum were obtained.By using moved overlapping separation ranging map(OSRM) and simulated chromatograms technology,the LC optimization method was developed. |
