助阳通络方治疗大鼠乳腺增生病的实验研究

摘    要:目的:利用动物模型,观察助阳通络方不同剂量组对乳腺增生病模型大鼠性激素水平的影响以及雌、孕激素受体表达情况,探讨其作用机制。方法:采用放射免疫法测定血清雌二醇(E2)、孕酮(P)、催乳素(PRL)的激素水平;采用免疫组化法测定乳腺增生病模型大鼠雌激素受体(ER)、孕激素受体(PR)阳性表达。结果:在助阳通络方对乳腺增生病大鼠性激素水平的影响实验中,助阳通络方高剂量组(E组)与正常对照组(A组)比较,E2、P、PRL均有显著性差异(P0.01);与疾病模型组(B组)比较,E2、P、PRL均有显著性差异(P0.01)。助阳通络方中剂量组(F组)与A组比较,无显著性差异(P0.05);与B组比较E2、P、PRL均有显著性差异(P0.01)。助阳通络方低剂量组(G组)与A组比较,E2、P、PRL均有显著性差异(P0.01);与B组比较E2、P、PRL均有显著性差异(P0.01)。在助阳通络方对乳腺增生病大鼠ER、PR的影响实验中,E组与A组比较,无显著性差异(P0.05);与B组比较,无显著性差异(P0.05)。F组与A组比较,无显著性差异(P0.05);与B组比较ER、PR阳性表达显著降低(P0.01)。G组与A组比较,ER阳性表达显著升高(P0.01);PR阳性表达升高(P0.05);与B组比较,无显著性差异(P0.05)。结论:助阳通络方可降低造模大鼠乳腺组织中升高的E2、PRL的含量,升高造模大鼠乳腺组织中降低的P含量,与三苯氧胺作用类似。可使造模大鼠乳腺组织中ER、PR阳性表达受抑制,且作用与三苯氧胺类似。

Abstract:Objective: To observe the effects of ZHUYANGTONGLUO decoction of different dose groups on the rat's model group of hyperplasia of mammary gland sex hormones and the expressions of estrogen,progestogen receptor,utilize animal's model.To explore its mechanism in therapeutic regulating sex hormones.Methods: Testing blood serum sex hormones of estradiol(E2),progestone(P),prolactin(PRL) adopting actino-immunization.And testing the expressions of estrogen(ER) and progestogen(PR) receptor adopting immunohistochemical staining.Results: In experimentation of mammary gland sex hormones,ZHUYANG-TONGLUO decoction high dose group(E group) is compared with normal group(A group),and E2,P,PRL are significant(P 0.01);And it is compared with model group(B group),E2,P,PRL are significant(P 0.01).ZHUYANGTONGLUO decoction middle dose group(F group) is compared with A group,E2,P,PRLare not significant(P 0.05);And it is compared with B group,E2,P,PRL are significant(P 0.01).ZHUYANGTONGLUO decoction low dose group(G group) is compared with A group,E2,P,PRL are significant(P 0.01);And is compared with compares B group,E2,P,PRL are significant(P 0.01).In experimentation of the expressions of ER,PR,E group is compared with A group,the expressions of ER,PR are not significant(P 0.05);And it is compared with B group,the expressions of ER,PR are not significant (P 0.05).F group is compared with A group,the expressions of ER,PR are not significant(P 0.05);And it is compared with B group,the expressions of ER,PR are significant(P 0.01).G group is compared with A group,the expressions of ER are significant(P 0.01),and the expressions of PR are not significant (P 0.05);G group is compared with B group,the expressions of ER,PR are not significant(P 0.05).Conclusions: 1.ZHUYANGTONGLUO decoction can lower the increasing E2,PRL of mammary gland and increase the degrading P of mammary gland,and pharmacodynamic action of it and tamoxifen are approximate.2.ZHUYANGTONGLUO decoction can restrain the expressions of ER,PR,and pharmacodynamic action of it and tamoxifen are approximate.
