酶法提取石榴皮多酚工艺研究

摘    要:为研究酶法提取石榴皮中多酚的最佳工艺,采用单因素试验考察不同浓度的纤维素酶、果胶酶、复合酶(不同质量比的纤维素酶和果胶酶)、酶解时间、酶解温度及酶解液pH值对石榴皮多酚得率的影响,并运用二次通用旋转回归组合设计优化酶法提取石榴皮多酚的最佳工艺参数。试验结果表明,对石榴皮多酚得率影响次序依次为酶解时间>酶浓度>pH值>酶解温度。当复合酶(纤维素酶和果胶酶质量比为2:1)质量浓度为0.25mg/mL,酶解时间150min,酶解温度50℃,初始酶解液pH6.0时,多酚得率达(23.87±0.08)%(n=5),与理论计算值23.96%的相对误差仅0.376%。酶法提取石榴皮中多酚的提取率比溶剂浸提法高出16.84%。

Abstract:The technology of extracting polyphenol from pomegranate peel by enzymic treatment was studied.The single factor experiments including different concentrations of cellulase,pectinase and compound enzyme(different mass ratio of cellulase and pectinase),enzymolysis time,enzymolysis temperature and enzyme solution pH on the yield of polyphenol was conducted.The extraction parameters of pomegranate peel polyphenol was optimized by the quadratic general rotation combination design.The results showed that the effects of pomegranate peel polyphenols yield was:enzymolysis time>enzyme concentration>pH>enzymolysis temperature.The results indicated that the optimal extraction conditions for maximum yield of polyphenol were:the amount of compound enzyme 0.25 mg/mL(the mass ratio of cellulase to pectinase is 2:1(W/W)),treatment time 150 min at 50 ℃,the initial enzyme solution pH 6.0.Under the optimized conditions,the yield is(23.87±0.08)%(n=5),the experimental values agree well with the estimated value 23.96% with relative error 0.376%.Yield of polyphenol increased by 16.84% by using the optimum conditions,as compared with the traditional ethanol extraction.
