类风湿关节炎患者外周血髓源性抑制细胞的检测及与Th1/Th2细胞的相关性分析

摘    要:目的: 检测类风湿关节炎(rheumatoid arthritis,RA)患者外周血中髓源性抑制细胞(myeloid-derived suppressor cells, MDSCs)的水平,分析其与Th1/Th2细胞的相关性。 方法:选取确诊的RA患者36例,根据临床资料计算DAS28评分,将其分为活动组和稳定组;另选取10例健康者作为对照组。3组MDSCs、Th1及Th2细胞的比例用流式细胞术检测并进行比较。分析MDSCs与Th1/Th2细胞的相关性。结果: 活动组MDSCs的比例明显高于稳定组和对照组(P均<0.05);稳定组MDSCS的比例与对照组比较,差异无统计学意义(P>0.05)。经Spearman相关性分析,MDSCs与Th1细胞呈负相关(P<0.05);而与Th2细胞无相关性(P>0.05)。 结论: MDSCs在RA活动期显著增高,且与CD4+T细胞亚群关系密切,可作为辅助评估RA活动状态的指标。

Abstract:Objective: To investigate the frequency of myeloid-derived suppressor cells(MDSCs) in the peripheral blood from patients with rheumatoid arthritis(RA) and assess the correlations between MDSCs and Th1/Th2 cells. Methods: A total of 36 RA patients were enrolled in the study. Disease activity was evaluated by calculating the 28-joint Disease Activity Score(DAS28) based on clinical information. RA patients were divided into active group and inactive group according to the DAS28 score. The frequency of MDSCs and Th1/Th2 cells in peripheral blood from RA patients and healthy controls were determined by flow cytometry. Results: The frequency of MDSCs increased significantly in active group compared with both inactive group and control group(P<0.05). However, there was no significant difference in the frequency of MDSCs between inactive group and control group(P>0.05). The frequency of MDSCs was negatively correlated with the frequency of Th1 cells in peripheral blood, but not correlated with the frequency of Th2 cells by Spearman′s test. Conclusion: The frequency of MDSCs in peripheral blood  from RA patients is highly prevalent and closely related to helper T cells, which may become a valuable marker for the evaluation of RA.
