摘    要:目的探讨丝素蛋白(SF)与丝素蛋白/羟基磷灰石复合材料(SF/HA)的体外细胞相容性,为组织工程提供理想的生物支架材料。方法将小鼠的骨髓间充质干细胞(C3H10T1/2)接种于上述两种生物材料上,运用四唑盐比色试验(MTT法)检测细胞的增殖能力,利用倒置显微镜和扫描电镜观察细胞在材料上的生长形态。结果 C3H10T1/2在SF与SF/HA上都能很好地黏附、生长和增殖。SF、SF/HA和C3H10T1/2都有良好的细胞相容性。结论 3%SF、6%SF、600%SF/HA3组多孔材料的体外细胞相容性最好。

Abstract:Objective To explore the cytocompatibility of the silk fibroin(SF) and SF/hydroxyapatite(HA) with murine mesenchymal stem cell line C3H10T1/2 in vitro.Methods The adherence and growth of the C3H10T1/2 cells on SF and SF /HA was observed by inverted phase contrast microscope and scanning electron microscope.The proliferation of C3H10T1/2 cells on various scaffolds was detected by MTT assay in vitro.Results Both SF and SF /HA could support the adherence,growth and proliferation of C3H10T1/2 cells,which suggested that they have good cytocompatibility and could be used as an ideal biomaterial in tissue engineering.Conlusion 3% SF,6% SF and 600% SF/HA porous materials have the best cytocompatibility in vitro.
