Abstract:Objective To investigate the effect of upregulation of miR-23 on apoptosis, invasion and migration of uterine fibroid cells based on mitogen-activated protein kinase/human Wnt-3 a protein/mammalian rapamycin target/vascular endothelial growth factor(MAPK/Wnt3 a/mTOR/VEGF)signaling. Methods Uterine fibroid cells were cultured and divided into control group, down-regulated group and up-regulated group. Proliferation, apoptosis, invasion and migration ability of each group were detected, and the expression levels of miR-23, plasma renin activity(PRA), matrix metalloproteinase-2(MMP-2), E-cadherin, MAPK, Wnt3 a, mTOR and VEGF were detected. Results Compared with the control group, miR-23 expression was lower and PRA expression was higher in the down-regulated group, and miR-23 expression was higher and PRA expression was lower in the up-regulated group, with statistical significance(P<0.05). Compared with the control group, the apoptosis rate and proliferation inhibition rate were lower and the number of invading cells and migrating cells were higher in the down-regulated group, while the apoptosis rate and proliferation inhibition rate were higher and the number of invading cells and migrating cells were lower in the up-regulated group, with statistical significance(P<0.05). Compared with the control group, the relative expression levels of MAPK, Wnt3 a, mTOR, VEGF and MMP-2 were higher in the down-regulated group, and the relative expression levels of E-cadherin were lower in the up-regulated group, while the relative expression levels of MAPK, Wnt3 a, mTOR, VEGF and MMP-2 were lower, and the relative expression levels of E-cadherin were higher, the difference was statistically significant(P<0.05). Conclusions Upregulation of miR-23 expression in uterine fibroids can regulate protein expression of MAPK/Wnt3 a/mTOR/VEGF signaling pathway, and then regulate apoptosis, invasion and migration of uterine fibroids.
