苦参碱抗肿瘤作用及其机制的初步研究

摘    要:目的:研究中药苦参碱(Matrine)的抗肿瘤作用及其免疫学机制。方法:MTT比色法检测肿瘤细胞的生长抑制率;流式细胞术分析细胞周期的改变。建立荷瘤小鼠模型,观察小鼠肿瘤的生长情况,计算肿瘤生长抑制率;MTT法测定苦参碱对荷瘤小鼠PBMC体外增殖作用的影响;ELISA法检测苦参碱治疗后小鼠血清中IL-2和IL-12的活性。结果:苦参碱可显著抑制小鼠肿瘤细胞的体外分裂和增殖,且抑制作用呈浓度和时间依赖性,使G1期细胞增多,S期和G2/M期细胞减少,细胞增殖指数降低。苦参碱对小鼠实体瘤生长具有显著抑制作用,抑瘤率高达60.7%以上;明显抑制小鼠静止PBMC的体外增殖作用;不能提高荷瘤小鼠血清中IL-2和IL-12的含量。结论:苦参碱具有较强的抗肿瘤作用,体内体外却表现出一定的免疫抑制作用,提高机体的免疫功能不是苦参碱发挥抗癌活性的主要机制。

Abstract:Objective: To explore the inhibitory effects of matrine on tumor growth and the possible mechanisms related to immunological functions. Methods: The tumor cell growth repression rate was measured by MTT colorimetry. Flow cytometry was used to analyze the cell cycle of H22 cells. The H22 BALB/c xenograft tumor models were established. The anti-tumor effect of matrine was observed and tumor growth inhibitory rate(IR) was calculated. The effect of matrine on the proliferation of peripheral blood mononuclear cell(PBMC) separated from H22 tumor-bearing BALB/c was determined by MTT assay in vitro; the content of serum IL-2 and IL-12 were examined by ELISA assay. Results: Matrine inhibited proliferation of the H22 tumor cells in both time and concentration-dependent manners, with increased the cell numbers at G1 phase and decreased numbers at S and G/M phase showing the possible mechanism for lowering cell proliferation rate. The tumor inhibition rate of matrien came up to 60.7% in tumor-bearing mice. Matrine significantly suppressed the proliferative activity of PBMC from tumor-bearing mice. Furthermore, martine could not increase the contents of murine serum IL-2 and IL-12. Conclusion: Matrine could inhibit tumor cell growth directly but not exert the anti-tumor efficiency through reinforcing the immune function.
