花花柴组织培养与植株再生体系的优化

摘    要:【目的】探讨植物生长调节剂、外植体及NaCl等因子对花花柴愈伤组织诱导、分化与植株再生的影响,并优化花花柴的再生植株体系,建立直接从茎端诱导丛生芽的植株再生体系。【方法】对生长约为2个月的花花柴植株进行不同的处理。采用不同浓度组合的细胞分裂素6-BA(6-Benzylaminopurine)和生长素NAA(α-Naphthaleneacetic acid)的MS(Murashige and Skoog)培养基(0.8%(w/v)琼脂粉)。【结果】最佳的愈伤组织诱导培养基为MS+6-BA 1.2 mg/L+NAA 0.6 mg/L;愈伤组织分化的最佳培养基为MS+6-BA3.0 mg/L+NAA2.0 mg/L;从外植体直接诱导不定芽的最佳培养基是MS培养基;诱导根的最佳培养基是1/2MS+NAA mg/L。【结论】花花柴叶片是愈伤组织形成的最佳材料,NaCl胁迫对愈伤组织的诱导有一定的抑制作用。

Abstract:【Objective】The purpose of this program was to explore the effects of plant growth regulators, explants and NaCl on the inducement to the callus,differentiation and plantlet regeneration of Karelinia caspica and optimize the system of tissue culture and plantlet regeneration of Karelinia caspica.【Method】The two-month-plandets of Karelinia caspica were treated with different concentrations of 6-Benzylaminopurine andα-Naphthaleneacetic acid in the MS culture medium.【Result】The results showed that leaves were the best explants to induce callus,NaCl supplement could inhibit the formation of callus,and the fittest callus induction medium was MS+0.6 mg/L 6-BA+1.2 mg/L NAA,and the best medium for adventitious buds induction was MS+3.0 mg / L 6-BA+2.0 mg /L NAA while root regeneration medium was 1/2MS+NAA 0.6 mg/L.【Conclusion】The leaves of Karelinia caspica are the best explants forming callus.NaCl stress is able to inhibit the callus induction.
