RSF-1在胶质瘤组织中的表达及其与病人预后的关系

摘    要:目的 探讨染色质重塑因子1(RSF-1)在人脑胶质瘤中的表达及其与病人预后的关系。方法 采用免疫组织化学染色法和免疫印迹法检测2010年1月~2013年4月手术切除的121例胶质瘤组织和2017年6月~2018年9月颅脑损伤内减压术中切取的50例正常脑组织RSF-1的表达水平。胶质瘤病人随访时间截止至2019年4月。多因素Cox比例回归风险模型分析影响胶质瘤病人生存预后的因素。结果 免疫组化染色结果显示胶质瘤组织RSF-1高表达率(66.12%,80/121)明显高于正常脑组织(0%;P<0.001)。免疫印迹法检测结果显示,胶质瘤组织RSF-1蛋白表达水平明显高于正常脑组织(P<0.05),而且,随胶质瘤级别增高,RSF-1蛋白表达水平明显增高(P<0.05)。多因素Cox比例回归风险模型分析结果显示,术前KPS评分<80分、WHO分级为Ⅲ~Ⅳ级、术后未化疗和RSF-1高表达是胶质瘤病人总生存期和无进展生存期缩短的独立影响因素(P<0.05)。RSF-1高表达病人总体生存期中位数(M):13个月;四分位间距(IQR):11~26个月]较低表达者(M:26个月;IQR:20~36个月)明显缩短(P<0.05)。 RSF-1高表达病人无进展生存期(M:11个月;IQR:7~23个月)较低表达者(M:21个月;IQR:16~28个月)明显缩短(P<0.05)。结论 RSF-1高表达可能预示着胶质瘤病人的不良生存预后

Abstract:Objective To investigate the expression of chromatin remodeling factor 1 (remodeling and spacing factor 1) in glioma tissues and its relationship with the prognosis of patients with glioma. Methods The expression levels of RSF-1 in 121 glioma tissues which were obtained from glioma patients underwent micorsurgery from January 2010 to April 2013 and in 50 normal brain tissues which were obtained from patients with traumatic brain injury underwent decompression from June 2017 to September 2018 were detected by immunohistochemical staining and western blotting. The follow-up time of glioma patients was up to April 2019. Cox proportional hazard model analysis was used to analyze the risk factors for the survival prognosis of glioma patients. Results The high expression rate of RSF-1 in glioma tissues (66.12%, 80/121) was significantly higher than that (0%) in normal brain tissues (P<0.001) according to the results of immunohistochemical staining. The results of western blotting showed that the expression level of RSF-1 protein in glioma tissues was significantly higher than that in normal brain tissues (P<0.05), and the expression level of RSF-1 protein increased significantly as the grade of glioma increased (P<0.05). The multivariate Cox proportional regression risk model analysis showed that the preoperative KPS score <80 points, WHO grade Ⅲ~Ⅳ, no postoperative chemotherapy and high RSF-1 expression were the risk factors for the shortening of overall survival time and progression-free survival time of glioma patients (P<0.05). The median overall survival time and progression-free survival time were significantly shorter in patients with high RSF-1 expression than those with low RSF-1 expression (P<0.05). Conclusion The high expression of RSF-1 may predict the poor survival prognosis of glioma patients
