摘    要:目的:以高效液相色谱法测定甜菊叶中绿原酸含量,正交试验法优选甜菊叶中绿原酸的最佳提取工艺。方法:以乙醇浓度、超声时间、料液比为考察因素,采用绿原酸提取率为指标,进行正交试验。结果:甜菊叶中绿原酸的最佳提取工艺为乙醇浓度60%,超声时间10 min,料液比1∶12,在最佳提取工艺下,绿原酸的提取率是4.03%。结论:通过正交试验优选的甜菊叶中绿原酸提取工艺稳定可行,适用于工业化生产。

Abstract:Objective:To determine the content of chlorogenic acid from Tianjuye(Stevia Rebaudiana) by high performance liquid chromatography(HPLC) and optimize the extraction process of chlorogenic acid from Tianjuye by orthogonal test. Methods:Orthogonal test was carried out with ethanol concentration,ultrasonic time and material liquid ratio as factors,and chlorogenic acid extraction rate as index. Results:The optimum extraction technology of chlorogenic acid from Tianjuye was ethanol concentration of 60%,ultrasonic time of10 minutes,material liquid ratio of 1 ∶12. Under the optimum extraction technology,the extraction rate of chlorogenic acid was 4.03%. Conclusion:The extraction process of chlorogenic acid from Tianjuye optimized by orthogonal test is stable and feasible which is suitable for industrial production.
