RAB5A基因对大肠癌细胞侵袭能力的影响

目的研究RAB5A基因转染后对大肠癌细胞侵袭能力的影响.方法用脂质体将RAB5A反义寡核苷酸转染人大肠癌细胞LS174T,48h后transwell小室法观察其对细胞转移能力的影响.结果 RAB5A反义寡核苷酸转染后48h,transwell小室结果可见转染后大肠癌细胞LS174T的侵袭和运动能力明显低于对照组(P《0.01).结论 RAB5A在人大肠癌细胞的侵袭及转移特性的形成中具有重要的作用,RAB5A的反义分子能够有效地阻断该基因表达的翻译过程,在细胞内发挥预期作用.

The effect of RAB5A gene on the invasion of colorectal neoplasm cells

Objective To study the effect of RAB5A gene transfection on the invasion of colorectal neoplasm cells.Methods RAB5A antisense oligonucleotides were transformed into human colorectal cancer cells LS174T by liposome and 48h later cellular metastasis ability was assayed through transwell method.Results 48h later after transfection with RAB5A antisense oligonucleotides,invasion and motor abilities of LS174T cells were lower in transwell method than those of controls(P<0.01).Conclusion The over expression of RAB5A gene plays an important role in the formation of tumor invasion phenotype and metastatic nature of human colorectal cancer cells.Antisense RNA can interrupt the translation of RAB5A and exert the expect effect.
