藏红花浸出液通过CD95/CD95-L途径诱导神经母细胞瘤发生凋亡

摘    要:目的 研究在藏红花作用下人神经母细胞瘤的凋亡发生与CD95受体表达的情况。方法人神经母细胞瘤SK-N-SH细胞的体外培养建立以后,分别加入不同浓度的藏红花浸出液进行培养。采用细胞计数法测定细胞生长曲线。细胞经碘化丙啶(PI)染色,采用流式细胞仪技术测定细胞周期及凋亡的发生。藏红花作用24h后,CD95受体表达通过抗CD95荧光抗体标记后的流式细胞仪方法检测。结果 藏红花浸出液(10mg/ml)对SK-N-SH细胞有明显的抑制作用,并可使S期细胞明显减少。浓度为10mg/lml和1mg/ml藏红花浸出液作用后细胞凋亡的发生明显增加。CDl95受体在所有SK-N-SH细胞中均有表达,经藏红花作用后,其表达有不同程度上调。结论 藏红花对SK-N-SH细胞具有抑制作用,其机制可能与藏红花能够诱导SK-N-SH细胞发生凋亡有关,CD95/CD95-L途径参与介导了诱发发生凋亡的过程。

Abstract:Objective To study apoptosis and expres s ion of CD95 in SK-N-SH cell line of neuroblastoma induced by saffron extracts. Methods Cell viability was tested by cell counting. Cell apoptosis and cell cycle were detected via flow cytometry after propidium iodide staining of DNA. The expression of CD95-receptor was assessed by flow cytometr y after incubation with R-phycoerythrin-conjugated anti-CD95-antibody. Results Saffron extracts had apparent inhibition on SK-N -SH cells and can significantly block S phase. Significant apoptosis was detect ed in SK-N-SH cells after the addition of 10mg/ml and 1mg/ml saffron extracts. CD95 expression was found on all SK-N-SH cells. The up-regulation of CD95 ex pression was seen in each saffron group. Conclusions Saffron extracts have inhibition on SK-N-SH cells. When these cells treated with saffron extracts, apoptosis is induced and expression of CD95 is up-regulated. CD95/CD95-L pathway may be involved i n apoptosis in SK-N-SH cell line induced by saffron extracts.
