番茄红素对心肌细胞缺氧损伤的作用机制

摘    要:目的探讨番茄红素对心肌细胞沉默信息调节因子1(Sirtl)-自噬通路的作用。方法原代培养乳鼠心肌细胞予以无血清、无糖培养及缺氧干预,模拟心肌缺血,用番茄红素和Sirtl的抑制剂尼克酰胺干预。采用Westernblot法检测Sirtl、自噬相关基因6(Atg6)Beclinl及自噬相关基因8LC3蛋白的表达;RT—PCR技术检测心肌细胞Sirtl的mRNA表达。结果①在心肌缺氧中,番茄红素提高了细胞Sirtl蛋白和mRNA的表达水平;②番茄红素增加了心肌缺氧时自噬相关基因Beelinl和LC3蛋白的表达;③Sirtl的抑制剂尼克酰胺阻断了番茄红素对白噬蛋白的作用。结论番茄红素可能通过Sirtl激活自噬,从而减少心肌缺氧损伤。

The mechanism of Lycopene in neonatal rat cardiomyocytes under hypox- ic condition

Abstract:Objective To investigate the effect of lycopene on Sirtl-Autophagy pathway in cardiomyocytes under hypoxic condition. Methods The primarily cultured neonatal rat cardiomyocytes were incubated under the condition of hypoxia in glucose free plus serum deprivation DMEM and exposed to lycopene or/and NAM. The protein level of Sirtl,Atg6 (Beclinl) and autophagic marker, LC3 were tested by Western blotting.The mRNA of Sirtl was detected by Real time PCR. Results Lycopene increased the protein and mRNA expression of Sirtl, and up regulated expression of Beclinl and LC3 protein under hypoxic condition. Blocking Sirtl by NAM could significantly decrease the effect of lycopene on autophagy in cardiomyocytes under hypoxic condition.Conclusion Lycopene may reduce myocardial damage by enhancing Sirtl-autophagy pathway under hypoxic condition.
