四生汤口服液对体外辐射人淋巴细胞的保护作用

目的观察四生汤口服液对受辐射的人体外周血淋巴细胞的防护作用。方法采用人外周血体外培养法,设对照组和3个不同浓度四生汤口服液组（1%、2%和4%）,通过流式细胞仪检测辐射后淋巴细胞凋亡率,应用3H-胸腺嘧啶核苷（3H-TdR）掺入实验观察受辐射淋巴细胞DNA的辐射抗性,MTT法检测不同照射剂量以及不同时间给药后,对照组及不同浓度实验组淋巴细胞增殖活性。结果照射后2h,各浓度四生汤组淋巴细胞的活性均高于对照组（P〈0.05）,且随浓度升高而增强;照射后5h加入四生汤口服液,只有高浓度组可以增强细胞活性（P〈0.05）,照射后10h加入四生汤口服液,均无保护作用（P〉0.05）。先向培养的淋巴细胞中加入不同浓度的四生汤口服液,再以1Gy和3Gy 60Co照射,1%、2%组3H-TdR掺入量明显高于对照组（P〈0.01）;照射剂量超过6Gy时,各浓度组3H-TdR掺入量与对照组差异无统计学意义。3Gy照射后,各浓度组淋巴细胞凋亡率均显著低于对照组（P〈0.01）。结论四生汤口服液对人淋巴细胞有一定抗辐射作用,这种作用受辐射剂量和辐照时间的影响。

Decoction of Four Fresh Drugs protects human lymphocytes against in vitro radiation

Objective To observe the protective effect of Decoction of Four Fresh Drugs on in vitro irradiated human peripheral blood lymphocytes.Methods In vitro cultured human peripheral blood lymphocytes were divided into control group,and 3 groups with different concentrations of Decoction of Four Fresh Drugs（1%,2%,and 4%） as experimental groups.Apoptosis rate of lymphocytes was detected with a flow cytometer.Radiation resistance of lymphocyte DNA was observed by 3H-thymidine incorporation.Proliferating activity of lymphocytes in control and different experimental groups was determined by MTT assay.Results Two hours after irradiation,the activity of lymphocytes was higher in different experimental groups than in control（P0.05）,which increased with the concentration of Decoction of Four Fresh drugs.Five hours after irradiation,only high concentration of Decoction of Four Fresh drugs increased the activity of lymphocytes（P0.05）.However,10h after irradiation,Decoction of Four Fresh drugs did not increase the activity of lymphocytes.After irradiation with 1Gy and 3Gy of 60Co,1% and 2% Decoction of Four Fresh Drugs with 3H-TdR significantly improved the radiation resistance of lymphocyte DNA（P0.05）.After irradiation with over 6Gy of 60Co,no difference was found in protective effect of Decoction of Four Fresh Drugs between control and experimental groups.After irradiation with 3Gy of 60Co,the apoptosis rate of lymphocytes was higher in control group than in experimental groups（P0.01）.Conclusion Decoction of Four Fresh Drugs protects human lymphocytes against irradiation in a dose-and time-dependent manner.
