摘    要:目的 建立测定骨碎补中柚皮苷含量的高效液相色谱法并优化炮制骨碎补的提取纯化方法.方法 Phenomenex C18柱(250 mm×4.6 mm,5 μm),流动相为乙腈-水-冰醋酸(27:72:1).流速1.0 mL/min,测定波长283 nm,温度30℃.结果 骨碎补浸膏以水(10倍,1.5 h,2次)、60%乙醇(10倍,1.5 h,2次)为溶剂提取,测得柚皮苷的含量分别为0.54%和0.56%.结论 结合生产实际,选择水提醇沉.分离纯化选择加硫酸铵至饱和,乙酸乙酯(20倍,2次)萃取,蒸干.

Abstract:Objective To establish a HPLC method for determining the naringin content and to optimize the extraction and purification method of processing Rhizoma Drynariae.Methods The HPLC method was adopted to measure the naringin content in Rhizoma Drynariae with the Phenomenex C18 column,the mobile phase of acetonitrile-water-glacial acetic acid(27 ∶72 ∶1),the flow rate of 1.0 mL/min,the detection wavelength of 283 nm and the temperature of 30 ℃.Results Rhizoma Drynariae extract was extracted with water(10 times,1.5 h,twice) and 60%alcohol(10 times,1.5 h,twice) as solvent.The determined contents of naringin were 0.54%and 0.56%respectively.Conclusion Combined with production practice,aqueous extraction-alcohol precipitation may be selected.Separation and purification may select adding ammonium sulfate to saturation and extracting by ethyl acetate(20 times,twice),then drying by distillation.
