青蒿素衍生物抗癌机制探讨

摘    要:目的:探讨青蒿素衍生物诱导肿瘤细胞产生自由基,引导肿瘤细胞死亡的机制。方法测定小鼠肿瘤动物模型的瘤体铁含量、观察青蒿素衍生物诱导肿瘤细胞产生自由基、采用单细胞彗星电泳法观察自由基对肿瘤细胞DNA杀伤的效应。结果小鼠的H22瘤体的铁含量显著高于血和肌肉、肺组织铁含量, Lewis肺癌的实验结果相同。K562细胞经青蒿素衍生物作用,能测出微量的自由基。青蒿素衍生物作用K562细胞可引起DNA损伤并溢出细胞。结论青蒿素衍生物可在含铁量较高的肿瘤细胞内产生自由基,导致肿瘤细胞死亡。

Approach free radical of artemisinine derivatives for anticancer mechanism

Abstract:Objective To explore mechanism of artemisinine derivatives induced tumor of cytogenic free radical and loading to tumors cell death.Methods Iron-content for tumor body of mouse tumor of animal model was evaluated and the antipersonnel effect of tumor of cytogenic free radical for DNA with artemisinine derivatives by Mono-cell comet caraphoresis induced was studied.Results The iron-content of blood, muscle and lung is toper of tumor-body. artemisinine derivatives induced K562 tumor cell of cytogenic free radical and caused DNA damnification and flowed outward of cell envelope.Conclusion Artemisinine derivatives can induce tumor of cytogenic free radical to kill tumor.
