舒络粉针对脑缺血大鼠氨基酸及其受体mRNA表达的影响

摘    要:目的研究舒络粉针对脑缺血大鼠海马区氨基酸含量和兴奋性氨基酸受体mRNA表达的影响。方法采用线栓法制成脑缺血模型,按其神经行为学进行分组。应用高效液相检测海马区脑匀浆谷氨酸(Glu)、γ-氨基丁酸(GABA)、天冬氨酸(Asp)、甘氨酸(Gly)含量;应用RT—PCR法研究舒络粉针对Glu受体-NMDAR1 mRNA表达的影响。结果舒络粉针能降低Glu含量、升高GABA含量,下调NMDAR mRNA的表达:结论舒络粉针可以抑制兴奋性氨基酸(EAA)的过量堆积、增加抑制性氨基酸(IAA)的含量而发挥抗神经元损伤的作用,其部分机理是与下调NMDAR的mRNA的表达有关。

Abstract:Aim To investigate effects of shuluo powder injection on excitatory amino in cerebral ischemia rats. Methods The model of cerebral ischemia rats was produced by transitory occlusion of the left middle cerebral artery (MCAO) with nylon line. The six groups were based by neurological deficits scores. GIu,GABA, Asp, and Gly were inspected by HPLC. The expression of NMDAR1 mRNA was detected by RT-PCR. Results The results indicate that shuluo can increase the content of GABA and reduce the content of Glu. Also the over expression of NMDAR1 was reduced following treatment of shuluo. Conclusions The role of neuroprotective effect of shuluo powder relates to restraining the over-aggregation of EAA and increasing the content of IAA, whose mechanism may decline over expression of NMDAR1.
