肌幼虫可溶性抗原与ES抗原检测抗旋毛虫抗体效果的比较

摘    要:目的比较旋毛虫肌幼虫可溶性抗原(SAg)与ES抗原(ESAg)检测抗旋毛虫抗体的效果。方法应用SAg-ELISA和ESAg-ELISA对旋毛虫病患者血清、感染旋毛虫的小鼠血清及肉汁、其他寄生虫感染人及动物血清进行抗旋毛虫抗体的检测。结果SAg-ELISA和ESAg-ELISA对10例旋毛虫病患者血清检测时抗体阳性率均为100%,但对10份正常人血清检测时SAg-ELISA的背景较深,两种抗原的A值相比较具有显著性差异(P<0.05);应用两种抗原对旋毛虫感染小鼠和正常小鼠血清及肉汁的检测结果与对人血清的检测结果相似。SAg与肝吸虫、肺吸虫、血吸虫等寄生虫感染人及动物血清均有交叉反应,而ESAg仅与1例肺吸虫病患者血清有交叉反应。结论旋毛虫肌幼虫SAg和ESAg检测抗旋毛虫抗体的敏感性相同,但ESAg的特异性优于SAg;肉汁也可作为样本进行抗旋毛虫抗体的检测。

Abstract:Objective To compare the efficacy of soluble antigen(SAg) and excretory-secretory antigens(ESAg) of T.spiralis muscle larvae for detection of anti-Trichinella antibodies.Methods Indirect ELISA using T.spiralis muscle larval SAg and ESAg were used to detect anti-Trichinella antibodies of sera from patients with trichinellosis,sera and muscle juice from mice infected with T.spiralis and sera from human and animals infected with other parasites.Results The positive rate of antibody was 100% for the 10 cases with trichinellosis assayed by ELISA using muscle larval SAg and ESAg.However,the background of SAg-ELISA was higher when SAg and ESAg were used to detect 10 normal persons,the difference of A value between SAg and ESAg was significant(P<0.05).The results of two antigens for detection of sera and muscle juice from normal mice and the infected mice were similar with those of human sera.SAg had cross-reactivity with human and animal infected with C.sinensis,P.westermani,S.japonicum,etc.,but ESAg had cross-reactivity with only one case with paragonimiasis.Conclusion The sensitivity of SAg and ESAg of T.spiralis muscle larvae for detection of anti-Trichinella antibodies was the same,but the specificity of ESAg was superior to SAg.Muscle juice could be used as the samples for detection of anti-Trichinella antibodies.
