维生素C防治百草枯中毒大鼠肝损伤的实验研究

摘    要:目的观察百草枯(PQ)中毒大鼠肝脏损伤的病理变化,并用抗氧化剂维生素C(VC)干预,观察中毒大鼠肝组织中肿瘤坏死因子-a(TNF-a)、白介素-1β(IL-1β)、超氧化物歧化酶-1(SOD-1)及丙二醛(MDA)的表达变化,研究其在百草枯(PQ)所致肝损伤中的作用。方法 54只SD大鼠,随机分为3组:对照组、染毒组、VC干预组。对照组用生理盐水一次性灌胃;染毒组给予生理盐水稀释PQ按180 mg/kg一次性灌胃,VC干预组于灌药同时给服VC溶液按180 mg/kg,分别于6、24、72 h后将其处死,留取肝脏,观察肝组织病理改变,并采用免疫组化SP法检测肝组织中MDA、SOD-1、TNF-a和IL-1β的表达。结果染毒组大鼠肝组织中TNF-aI、L-1β及MDA的表达均高于对照组(P〈0.05),而VC干预组明显低于单纯染毒组(P〈0.05);染毒组SOD-1表达低于对照组(P〈0.05),而VC干预组明显高于单纯染毒组(P〈0.05)。结论 MDA、SOD-1、TNF-a和IL-1β在PQ中毒大鼠肝损伤中起重要的调节作用,VC可下调肝组织中促炎性细胞因子的表达,提高组织SOD活性,对改善PQ中毒大鼠肝细胞抗氧化能力有积极作用。

Abstract:Objective To study the pathological changes and the expression of MDA,SOD-1,TNF-a and IL-1β in liver of rats with acute paraquat poisoning and the role of the antioxidant vitamin C in PQ-induced liver injury.Methods 54 SD rats were randomly divided into control group,poisoned group and VC intervention group.Gastric was perfused with paraquat 180 mg/kg once.Meanwhile,intervention group were perfused with VC 180 mg/kg.At 6 h,24 h,72 h after exposure,the animals in each group were used to evaluate lung pathological changes and the expression of MDA,SOD-1,TNF-a and IL-1β in livers by Immunohistochemistry(IHC).Results When compared with control group,the expression of MDA,TNF-a and IL-1β in intoxication group was significantly higher(P<0.05).Moreover,expression of SOD-1 was lower than the control group(P<0.05).The levels of MDA,TNF-a and IL-1β decreased in VC intervention group when compared with PQ group(P<0.05) and the SOD vitality increased markedly when compared with PQ group(P<0.05).Conclusion MDA,SOD-1,TNF-a and IL-1β may play an important role in liver injury of poisoned rats.Vitamin C could act as an immunology regulator,anti-inlfammatory factor and improve the hepatic antioxidant ability.
