摘    要:目的应用酶标法检测干扰素脂质体包封率。方法应用ELISA法测定干扰素脂质体裂解前后的干扰素含量,计算脂质体的包封率,并与传统的Wish细胞病毒抑制法比较。结果ELISA法在干扰素活性为104~107IU/ml范围内,能检出的结果精密度明显高于传统Wish细胞病毒抑制法。Wish细胞病毒抑制法测定的3批样品包封率的CV值分别为8·6%、11·2%和12·70%,ELISA法测定的CV值分别为3·1%、4·3%和4·7%,二者差异有显著意义。结论ELISA法可以快速、高通量、准确地检测干扰素脂质体的包封率。

Abstract:Objective To develop a novel method for determination of entrapment efficiency of interferon liposome.MethodsDetermine the interferon contents in interferon liposome before and after splitting,and calculate the entrapment efficiency.Compare the result with that determined by Wish cell virus inhibition test.Results By using ELISA,the interferon activity ranged from 104 to 107 IU/ml was detected.The accuracy of ELISA was significantly higher than that of Wish cell virus inhibition test.The CV values of entrapment efficiencies of 3 lots of interferon liposome determined by Wish cell virus inhibition test were 8.6%,11.2% and 12.70% respectively,which were significantly different from those by ELISA(3.1%,4.3% and 4.7% respectively).Conclusion Compared with traditional Wish cell virus inhibition test,ELISA was more rapid,accurate and of a high throughput in determination of entrapment efficiency of interferon liposome.
