Abstract:OBJECTIVE To study the role of recombination activating gene 1 (RAG1) in pathogenesis of allergic rhinitis, and to analyze the regulative effect of dexamethasone (DEX) on RAG1. METHODS Rat model was established by ovalbumin (OVA) challenge method. Passive cutaneous anaphylaxis (PCA) and counting sneeze test was performed in three groups and the serum levels of IgE were determined by ELISA method. The expression and DNA sequence of recombination activating gene 1 in rats' thymus were analyzed by RT-PCR and DNA analysator. RESULTS The sneezing time of rats in AR group was significantly higher than that in other 2 groups (P<0.05) . The positive rate of PCA test was 90 % in AR group and was negative in the other 2 groups. Serum level of IgE in AR group increased markedly compared with that in normal control group. In AR DEX group, serum level of IgE decreased significantly compared with that in AR group. The semi-quantitation of RAG1 in normal control group was significantly lower than that in AR group and in AR DEX group (P<0.05) . However, the difference of semi-quantitation between AR group and AR DEX group was not significant (P> 0.05) and the homology of DNA sequence was 100 % between determine sequence and known sequence. CONCLUSION RAG1 may relate with allergic rhinitis but without mutation and deletion on the pathogenesis of allergic rhinitis. Glucocorticoid is an effective immunomodulator but cannot change the expression of RAG1, so RAG1 may be a higher immunomodulator.
