黑曲霉 G-1125生淀粉糖化酶基因的克隆与序列分析

摘    要:从黑曲霉Aspergillus niger G-1125克隆到了生淀粉糖化酶菌基因的DNA及cDNA序列。序列分析表明,该生淀粉糖化酶基因组DNA 序列编码区长2 172 bp,cDNA编码区长1 923 bp,该基因含有4个内含子,共编码640个氨基酸,前18个氨基酸为信号肽序列,该氨基酸序列中共含有2个潜在的糖基化位点,软件预测出该酶的分子质量约为68 ku,等电点为pH值4.07。

Abstract:DNA and cDNA of raw-starch-digesting-glucoamylase were cloned from Aspergillus niger G-1125. The sequence analysis revealed that the length of DNA and cDNA of this gene were 2 172 bp and 123 bp, respectively. The DNA sequence contained 4 introns and encoded a protein of 640 amino acids with a signal peptide of 18 amino acids and two potential glycosylation sites. The calculated molecular weight and isoelectric point of the enzyme were 68 ku and pH 4.07, respectively.
