摘    要:目的]了解呼吸道合胞病毒(RsV)感染对鼻黏膜下成纤维细胞分泌趋化因子的影响.方法]取手术切除的下鼻甲鼻黏膜进行上皮细胞及黏膜下成纤维细胞培养,将RSV感染鼻黏膜上皮细胞后,用其培养上清液刺激培养的成纤维细胞,用荧光定量PCR分别于6,12,24,48h时测定嗜酸细胞活化趋化因子、调节活化正常T细胞表达和分泌的趋化因子(RANTES)、白细胞介素8(IL-8)及生长调节癌基因α(GRO—α)的mRNA表达程度;用ELISA方法分别于24,48h测定培养液中嗜酸细胞活化趋化因子、RANTES,IL-8,GRO-α蛋白质的含量.结果]实验组IL-8,GRO-αmRNA的表达程度明显高于对照组,RANTES,IL-8,GRO-α蛋白表达量明显高于对照组.结论]被RSV感染的鼻黏膜组织中成纤维细胞有

Abstract:OBJECTIVE To study the effect of respiratory syncytial virus (RSV) infection on chemokine secretion of fibroblast under nasal mucosa. METHODS Epithelial cells and fibroblasts under nasal mucosa from surgical removal of nasal mucosa of inferior turbinate were cultured, and the fibroblasts were cultured with the supernatant fluid of nasal mucosa epithelial cells infected with RSV by co-cultivation. The expression of mRNA and protein level of eotaxin, regulated upon activation normal T cell expressed and secreted (RANTES), IL-8 and growth-regulated protein alpha (GRO-a) were measured in culture solution with realtime PCR at 6, 12, 24, 48 h and detected by ELISA at 24 and 48 h. RESULTS The expression of IL-8 and GRO-α mRNA, and the protein level of RANTES, IL-8 and GRO-α were significantly higher in experiment group than in control group. CONCLUSION Fibroblast under nasal mucosa infected by RSV has demonstrably secretory role of cytokins.
