人WDR70蛋白结构与功能的生物信息学预测

摘    要:人WDR70蛋白作为一个衔接蛋白,可介导多蛋白相互作用,参与DNA损伤修复、胚胎发育等生物过程。采用生物信息学软件对人WDR70蛋白的结构和功能进行分析,发现WDR70蛋白的相对分子质量约为73 kD,pI为5.94,半衰期为30h,不含有信号肽和跨膜区结构;同源蛋白的多重序列比对和进化分析显示,人WDR70蛋白与黑猩猩、白枕白眉猴、食蟹猕猴的WDR70蛋白具有高度的同源性,并且在系统发育树上聚为一类;无规则卷曲是WDR70蛋白二级结构主要的结构形式,三级结构的预测结果与之吻合;功能分类预测其为结合蛋白,参与蛋白的泛素化和转运过程。开展WDR70蛋白的生物信息学分析,为了解该蛋白的结构和功能提供信息依据,为后续WDR70功能研究提供新思路。

Abstract:As a adaptor protein, human WDR70 protein mediated the interaction of multi-proteins and participated in the biological process, such as DNA repairation and embryonic development etc. The analysis results by bioinformatic sofwares showed that WDR70 was a hydrophilic protein, the molecular mass was about 73 kD, its theoretical isoelectric point and half-life were 5.94 and 30 hours respectively. The protein has no signal peptide and transmembrane regions. The multiple alignment and clustering analysis showed that human WDR70 was highly homologous with that of Pan troglodytes, Cercocebus atys and Macaca fascicularis. The random coil was the main structure form in the WDR70 protein secondary structure. The three-demensional structure prediction expressed similar results as secondary structure prediction. Functional classification prediction showed that WDR70 was binding protein which involved in the protein transport and ubiquitination. The bioinformatics analysis of WDR70 protein would provide information basis for understanding the structure and function of the protein, and provide new ideas for further research on WDR70 function.
