宫颈黏膜细胞线粒体DNA核内整合及其与c-myc基因表达的关系

摘    要:目的:探讨宫颈黏膜细胞核内线粒体DNA(mtDNA)整合与c—myc基因表达的关系。方法:采用免疫组(IHC)检测c—myc基因表达:采用原位杂交方法对宫颈黏膜细胞核内mtDNA序列进行检测:结果:宫颈癌黏膜细胞核内mtDNA序列的检出率为27.5%,不典型增生为13.3%、正常宫颈黏膜细胞未发现有,mtDNA序列整合 宫颈黏膜细胞c-myc基因表达率在mtDNA序列检出组为67%,显著高于mtDNA序列列未检出组(36%)。结论:宫颈黏膜细胞mtDNA序列核内整合可能参与了宫颈癌的发生:c-myc基因表达可能与宫颈黏膜mtDNA序列整合有关。

Abstract:Objective: To explore the relationship between the integration of mtDNA in the nuclei of cervical epithelium cells and the expression of c-myc. Methods:The expression of c-myc proteins was examined in 40 cases of uterine cervix carcinoma and 30 cases of CIN and 30 cases of cervical epithelium; the sequence of mtDNA in the nuclei was detected by in situ hybridization. Results:The integration rate of mtDNA in the nuclei was 27.5 % in cervical carcinoma,13.3% in CIN, 0% in cervical epithelium. The expression rate of c-myc in integration group(67%)was significantly higher than that in integration- group (36%)?Conclusion:Integration of mtDNA into nuclei of cervical epithelium cells may be involved in the carcinogenesis of cervical epithelium cells and may contribute to the expression of c-myc.
