Abstract:Objective In order to investigate the mechanism of Piyan Yihao Fang in treatment of urticaria based on network pharmacology.Methods Traditional Chinese medicine systems pharmacology(TCMSP)was used to screen the active compounds and targets of Piyan Yihao Fang.The related targets of urticaria were found in GeneCard,Online Mendelian Inheritance in Man(OMIM),PharmGKB and Therapeutic Target Database(TTD).The"drug-common target-disease"network was constructed,and the interaction between common targets was found by constructing the Protein-Protein Interaction Network(PPI).Gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed on the common targets.Results TCMSP database indicated that there were 124 active compounds and 134 targets of Piyan Yihao Fang.GeneCards database indicated that there were 999 targets closely related to urticaria,and there were another 36 common targets for drugs and diseases.Venn diagram showed that there were 36 common targets of Piyan Yihao Fang and urticaria.GO function enrichment analysis showed that 1243 items were involved in the common target,including 1083 biological processes(BP),58 cellular components(CC)and 102 molecular functions(MF).Its biological process mainly concentrated in hypoxia response,response to glucocorticoid,corticosteroid response,G protein-coupled acetylcholine receptor signaling pathway,response to decreased oxygen levels,etc.KEGG pathway enrichment analysis suggested that intersection targets involved 96 signaling pathways,mainly covered the prostate cancer,kaposi sarcoma-associated herpesvirus infection,advanced,glycation,endproduct-receptor of advanced glycation endproduct(AGE-RAGE)signal pathway in diabetic complications,phosphatidylinositol 3-kinase(PI3K)-protein kinase B(AKT)signaling pathway,Platinum drug resistance,etc.The results of molecular docking showed that there was a good binding force between the main compounds and the main targets.Conclusion The treatment of urticaria with Piyan Yihao Fang involves multiple signal pathways and biological processes,which also reflects the treatment characteristics of traditional Chinese medicine with multiple targets and multiple pathways.
