蜂毒肽对大鼠慢性前列腺炎疼痛的作用

摘    要:目的 观察蜂毒肽(melittin)对大鼠前列腺炎疼痛模型的治疗作用.方法 通过手术将弗氏完全佐剂(CFA)注射到SD大鼠前列腺中建立模型,继续饲养12 d后再次通过手术将蜂毒肽注射到大鼠前列腺内进行干预.在注射CFA后的第6、12、18天进行机械痛阈测定,然后取前列腺和脊髓标本分别进行组织学、环氧合酶-2(COX-2)以及脊髓背角胶质原纤维酸性蛋白(GFAP)的测定.对照组大鼠,前列腺内注射等量生理盐水,饲养相同时间后同方法检测.结果 CFA诱导的大鼠前列腺炎模型能产生痛觉过敏,前列腺内炎症细胞数量和COX-2表达均增加,脊髓中GFAP表达增加.同对照组比较,蜂毒肽注射后大鼠机械痛压力阈值提高,前列腺中炎症细胞数量和COX-2表达均降低,脊髓中GFAP表达降低.结论 蜂毒肽注射能够有效提高前列腺炎大鼠的痛阈,抑制炎症细胞、COX-2和GFAP的表达,发挥镇痛和抗炎作用.

Abstract:Objective To test the effectiveness of melittin in chronic prostatic pain model. Methods Adult male Spraque-Dawley rats were injected with complete freunds adjuvant (CFA) into the prostate to make chronic prostatic inflammation. Melittin was injected into the prostates of these models 12 days later. The nociceptive effects were evaluated by using a behavior approach ( mechanical threshold measurement) at the 6th, 12th, and 18th day respectively after intraprostatic injection of CFA. The level of cyclooxygenase (COX-2) in prostate and glial fibrillary acidic protein (GFAP) expression in spinal cord were detected by immunohistochemistry. The rats in control group were injected with normal saline. Results CFA induced rats local allodynia, increases in inflammatory cell quantity and COX-2 expression in prostate, and GFAP expression in spinal cord. Melittin injection significantly raised pain mechanical threshold and decreased inflammatory cells quantity, COX-2 and GFAP expression as compared with control group. Conclusion Local injection of melittin can raise pain threshold, inhibit accumulation of inflammatory cells and expression of COX-2 and GFAP, produce some analgesic and anti-inflammatory effects in rat chronic prostate pain model.
