激素和外植体对亚麻愈伤组织形成的影响

摘    要:为了建立亚麻的愈伤组织再生体系,以亚麻无菌苗的下胚轴、子叶为外植体,研究了不同激素、外植体对亚麻愈伤组织形成的影响。结果显示:亚麻种子最佳的灭菌方法是用0.1%的HgCl2处理6 min;亚麻愈伤组织适宜形成外植体的部位是下胚轴,可以形成较多疏松、绿色的愈伤组织;4 mg/L浓度的6-BA有利于子叶愈伤组织的形成,愈伤组织的形成百分率最大,为87.5%;子叶诱导愈伤组织的最佳配方为MS+4 mg/L 6-BA+0.05 mg/L IAA;1~4 mg/L 6-BA+0.05 mg/L IAA处理亚麻下胚轴均可有效诱导其愈伤组织的分化。

Abstract:The impact of hormone and explants on callus of flax were researched with hypocotyls and cotyledon of germfree seedlings in order to set up callus regeneration system. The results show that the best method of seed sterilization was the treatment of 6 min by 0.1% HgCl2. Then the best explants were hypocotyls for more green loose callus. The 4 mg/L 6--BA promoted more callus production of cotyledon with the most callus of 87.5%. The best culture medium for cotyledon callus production was MS+4 mg/L 6-BA+0.05 mg/L IAA, and the best one for hypocotyls callus was 1-4 mg/L 6-BA+0.05 mg/L IAA.
